Production Procedures

Antibody was affinity purified using an epitope specific to HMGN1 immobilized on solid support.

The epitope recognized by A302-363A maps to a region between residue 90 and 100 of human high-mobility group nucleosome binding domain 1 using the numbering given in entry NP_004956.5 (GeneID 3150).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

Antibody was affinity purified using an epitope specific to HMGN1 immobilized on solid support.

HMGN1 and its close analog HMG17 (MIM 163910) bind to the inner side of the nucleosomal DNA to potentially alter the interaction between the DNA and the histone octamer. The two proteins may be involved in the process that maintains transcribable genes in a unique chromatin conformation. Their ubiquitous distribution and relative abundance, as well as the high evolutionary conservation of the DNA-binding domain of the HMG14 family of proteins, suggest that they may be involved in an important cellular function [taken from NCBI Entrez Gene (GeneID:3150)].

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of cell lysates are performed using Goat anti-Rabbit IgG Heavy and Light Chain Antibody (Cat. No. A120-101P).
