Antibody was affinity purified using an epitope specific to DFF45/DFFA immobilized on solid support.

The epitope recognized by IHC-00291 maps to a region between residue 271 and 331 of human DNA fragmentation factor, 45kDa, alpha polypeptide using the numbering given in entry NP_004392.1 (GeneID 1676).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

DFF (DNA fragmentation factor) is the principle endonuclease responsible for DNA cleavage during apoptosis and exists as a heterodimer formed between the inhibitory subunit, DFF45, and the DNA nuclease, DFF40. DFF35/DFFA is a 35 kDa splice variant of the inhibitory subunit, DFF45. Similar to DFF45, DFF35 has been shown to heterodimerize with DFF40 and also negatively regulate its nuclease activity. In addition to regulating DFF40 activity, DFF35 has also been found to assist in the synthesis and folding of the DFF40 endonuclease.
