Antibody was affinity purified using an epitope specific to CAD immobilized on solid support.
The epitope recognized by IHC-00280 maps to a region between residue 1650 and 1700 of human carbamoyl-phosphate synthetase 2, aspartate transcarbamylase, and dihydroorotase using the numbering given in entry NP_004332.2 (GeneID 790).
Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

CAD is an enzyme involved in the first three steps of the de novo synthesis of pyrimidine nucleotides. CAD exhibits multiple catalytic activities and is described as a “fusion” protein that can function as a carbamoylphosphate synthetase, aspartate transcarbamoylase, and dihydroorotase. CAD has been shown to be phosphorylated by mitogen-activated protein (MAP) kinase and protein kinase A (PKA) for the coordination of cell cycle progression and pyrimidine biosynthesis.
