p53, the prototypical tumor suppressor, is a critical intracellular translator of extracellular signals such as UV- or gamma-radiation, nutrient depravation, or genotoxic drugs. In coordination with the regulatory effects of other proteins such as HdmX, p53 activation regulates a huge number of genes and results in cell cycle arrest, inhibition of angiogenesis, and a number of other effects. Mutated in over half of human cancers, understanding this protein and its pathways will have a significant impact on human health.
