Antibody was affinity purified using an epitope specific to MAP1A immobilized on solid support.

The epitope recognized by A301-444A-T maps to a region between residue 625 and 675 of human microtubule-associated protein 1A using the numbering given in entry NP_002364.5 (GeneID 4130).

Immunoglobulin concentration was determined using Beer’s Law where 1mg/mL IgG has an A280 of 1.4.

MAP1A is one of the founding members of the MAP (microtubule-associated protein) family of proteins involved in microtubule assembly. MAP1A is almost exclusively expressed in the mature brain and is important to dendrite formation. MAP1A is processed by proteolytic cleavage that leads to the generation of a MAP1A heavy chain of 350 kDa and a LC2 light chain of 28 kDa. Both the heavy and light chain have been shown to bind microtubules as well as a number of other proteins that play a role in synapse formation.

Applications

All western blot analysis is performed using 5% Milk-TBST for blocking and as antibody diluent. Primary antibody is incubated overnight.

Western blots of immunoprecipitates are performed using Goat anti-Rabbit Light Chain HRP Conjugate (Cat. No. A120-113P) with 5% Normal Pig Serum (Cat. No. S100-020) added to the blocking buffer.
