Detection of Human and Mouse PPP2R5D by Western Blot (h&m) and Immunoprecipitation (h). Samples: Whole cell lysate from HeLa (5, 15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 µg) and mouse NIH3T3 (M; 50 µg) cells. Antibodies: Affinity purified rabbit anti-PPP2R5D antibody A301-100A used for WB at 0.04 µg/ml (A) and 0.1 µg/ml (B) and used for IP at 3 µg/mg lysate. PPP2R5D was also immunoprecipitated by rabbit anti-PPP2R5D antibodies A301-098A and A301-099A, which recognize upstream epitopes. Detection: Chemiluminescence with exposure times of 30 seconds (A and B).

PP2A is a major serine/threonine phosphatase that is implicated in the negative control of cell cycle progression. It is a trimeric holoenzyme that is comprised of a catalytic, structural, and regulatory subunit. PPP2R5D is a regulatory subunit of PP2A. The regulatory subunits of PP2A are grouped into 3 families: B/PR55, B’/PR61, and B’’/PR72. PPP2R5D encodes the delta isoform of the B’ subunit, B56.
