Actin Cytoskeleton

The actin cytoskeleton plays a key role in several cell functions such as cell polarization, migration, endocytosis, and intracellular transport. The actin cytoskeleton is composed of actin monomers (G-actin) that polymerize into actin filaments (F-actin) in response to extracellular stimuli and intracellular cues. Rho GTPases and phosphoinositide lipids stimulate the formation of actin structures such as lamellipodia (Rac GTPase), filopodia (Cdc42), and stress fibers (Rho) (1). The timing and localization of actin polymerization is regulated by actin polymerization machinery such as the WASP-Arp2/3 complex, which nucleates branched filament polymerization and formins, which nucleate unbranched actin filaments (2). The stability of polymerized actin is dynamically controlled by actin-binding proteins, which fine-tune the balance between actin depolymerization and actin nucleation, influence actin filament length, modulate actin filament assembly and turnover, and cross-link filaments into higher order actin structures (2). Finally, focal adhesions contain proteins, such as Cortactin, that bind actin and anchor the actin cytoskeleton to the extra cellular matrix (3). Because regulation of the actin cytoskeleton is involved in the processes of cell polarization and migration, study of the actin cytoskeleton is critical to understand aspects of human disease such as T-cell polarization and activation in response to antigen presenting cells (4) and cancer cell migration and invasion (5).
