SOS1 (son of sevenless homolog 1) is a human paralog of the drosophila SOS gene. Drosophila SOS was originally identified as a gene that functioned downstream of the sevenless gene in the Ras/MAP kinase signaling pathway. SOS and its paralogs, SOS1 and SOS2, function as guanine nucleotide exchange factors that act on Ras-GTPases to cause release of GDP in exchange for GTP. Mutations in SOS1 are the cause of Noonan syndrome type 4 and gingival fibromatosis.
